摘要
目的建立HPLC-ELSD法测定降糖胶囊中菝葜皂苷元的含量方法。方法采用HPLC-ELSD法测定降糖胶囊中菝葜皂苷元的含量。采用乙醇提取样品,SUPELCO Discovery C_(18)柱(250mm×4.6mm,5μm),以甲醇-水(90∶10)为流动相,流速1.0m L·min^(- 1),柱温为30℃,蒸发光散射检测器漂移管温度为45℃,以氮气为雾化气,压力为486k Pa,增益倍数为6。结果含量测定中菝葜皂苷元在0.05243~0.2996mg·m L^(- 1)浓度范围内呈良好的线性关系(r=0.9999)。结论建立的分析方法简便可行,专属性强,可用于降糖胶囊中菝葜皂苷元的定量检测。
OBJECTIVE HPLC method for determination of sarsasapogenin in Jiangtang capsule by HPLCELSD method. METHODS Determination of sarsasapogenin in Jiangtang capsule by HPLC-ELSD. The samples was extracted with ethanol,SUPELCO Discovery C_（18） column（ 250 mm × 4. 6mm,5μm）,methanol∶ water（ 90∶ 10） as mobile phase,flow rate 1. 0m L·min^（-1）,column temperature was 30℃,the ELSD drift tube temperature of 45℃,using nitrogen as the atomization gas,pressure was 486 k Pa,gain of 6. RESULTS The content determination of sarsasapogenin in 0. 05243 mg · m L^（-1）~ 0. 2996 mg · m L^（-1） concentration range showed a good linear relationship（ r =0. 9999）. CONCLUSION The established method is simple,reproducible,and can be used for the quantitative determination of in Jiangtang capsule.
