摘要
目的 研究有关生化因素对喜树碱(CPT)诱导人鼻咽癌细胞凋亡的影响。方法 在CPT诱导人鼻咽癌CNE-2Z 细胞凋亡的基础上,采用流式细胞仪及二苯胺反应等方法,观察了多胺、Ca2+、Mg2十依赖性内切酶活性抑制剂及PARP活性抑 制剂对DNA片段化形成的影响。结果 多胺可明显抑制CPT诱导的CNE-2Z细胞凋亡,使片段化DNA明显降低,P<0.05。内 切酶及PARP活性抑制剂与CPT共同作用CNE-2Z细胞后测得片段化DNA与单用CPT相比无显著性差异,P>0.05。结论 喜树碱诱导的 CNE-2Z细胞凋亡可能与一种非 Ca2+、Mg2+依赖性内切酶活性有关,且受多胺的调节。
Objective To study the effects of some biochemical factors on apoptosis in nasopharyngeal carcinoma cells (CEN-2Z) induced by DNA topoisomerase I inhibitor camptothecin (CPT). Methods DNA fragmentation was analysed with diphenylamine reaction. Results Spermine could obviously reduce the DNA fragmentation in CHE-2Z cells beated with 4 mol/L CPT, P<0.05. Neither depletion of intracellular calcium by EGTA nor inhibiting the activity of an endonuclease or poly (ADP-ribose) polymerase could effect the DNA fragmentation in CNE-2Z cells treated with 4 mol/L CPT, P>0.05. Conclusion The above results suggested that a Ca2+.Mg2+ independent endonuclease might be involved in the apoptosis in CNE-2Z cells induced by DNA topoisomerase I inhibitor camptothecin.
