摘要
目的 研究人参皂苷 (GRS)对神经元分化细胞株PC12损伤及凋亡的影响。方法 采用神经细胞培养和流式细胞仪技术 ,以 β淀粉样蛋白 (Aβ)诱导PC12细胞损伤作为细胞模型 ,观察Aβ对PC12细胞的影响以及不同剂量GRS对PC12损伤的保护作用。结果 CRS 5 ,10 μg·ml-1明显减少Aβ介导PC12损伤时细胞内酶的释放 ,提高细胞的存活率。对Aβ诱导的细胞凋亡 ,GRS也有一定的拮抗作用。结论 人参皂苷能拮抗Aβ诱导的神经细胞凋亡 ,对Aβ引起的神经细胞毒性有一定的保护作用。
OBJECTIVE To study the effects of ginsenoside (GRS) on injury and apoptosis of pheochromocytoma (PC12) cells induced by β amyloid peptide (Aβ).METHODS Pheochromocytoma(PC12) cells induced by Aβ were used to observe the effects of GRS on the inhibition of apoptosis.Apoptotic cells were measured using FACS analysis after staining with Annexin V fluos(FL1) and propidium.RESULTS 5 and 10 μg·ml -1 of GRS attenuated neuronal damage induced by Aβ,and decreased of enzyme release from PC12 .GRS inhibited Aβ induces apoptosis of cultures PC12 cells.CONCLUSION Aβ(5 μg·ml -1 )induced apoptosis in cultured PC12 cells and GRS(5~10 μg·ml -1 ) reduced apoptosis.
