摘要
目的:探讨大鼠丹防胶囊含药血清对肝星状细胞合成血小板源性生长因子-B(PDGF-B)及细胞外信号调节激酶1/2(ERK1/2)的影响。方法:将大鼠分为3组,分别经丹防胶囊、复方鳖甲软肝片及生理盐水灌胃后股动脉取血制备大鼠丹防胶囊含药血清、鳖甲含药血清及生理盐水血清,3组血清分别干预肝星状细胞(HSCT6),免疫细胞化学法、蛋白印迹法检测PDGF-B及ERK1/2表达的水平。结果:丹防胶囊血清组HSC-T6中PDGF-B及ERK1/2蛋白表达强度均较生理盐水血清组明显降低(P<0.01),与鳖甲血清组相当(P>0.05)。结论:丹防胶囊大鼠含药血清对肝星状细胞合成PDGF-B及ERK1/2有明显的抑制作用,提示丹防胶囊可能对大鼠肝星状细胞的增殖具有抑制效应。
Objective： To investigate the effect of Dan-fang Capsule on the expression of platelet de- rived growth factor B （PDGF-B） and extracellular signal-regulated kinase （ERK） 1/2 in hepatic stellate cells （HSC）. Methods： Serum of Dan-fang capsule treated rats was ursed to intervene the activity of PDGF-B, ERK1/2 in HSC-T6, and immunocytochemistry method and western blot were used to de- termine the expression of PDGF-B and ERK1/2 in HSC lysates. Results： Expression of PDGF-B and ERK1/2 were significantly lower in rats of Dan-fang capsule treated group than in rats of control group （P 〈0.01 ）. Conclusion： Serum of Dan-fang capsule treated rats showed obvious inhibiting effect on PDGF-B and ERK1/2 in HSC, which suggests Dan-fang capsule might have inhibiting effect to proli- feration of HSC.
