摘要
目的:研究大鼠尿毒症血清干预对人脐静脉内皮细胞(HUVEC)的作用及临床意义。方法:将20只SD大鼠随机分为健康组(假手术组)和尿毒症12周组。对尿毒症组大鼠进行5/6肾切除,致其慢性肾衰。以不同体积分数(10%、15%、20%、25%)的尿毒症和健康大鼠血清处理HUVEC 24h,显微镜下观察HUVEC形态改变,MTT法测细胞活力,TBA法测胞浆丙二醛含量,RT-PCR法测定IL-6、TNF-α、NF-κB、Caspase-3mRNA相对表达量。结果:尿毒症环境能使细胞活力降低,使细胞内MDA、IL-6、TNF-α、NF-κB、Caspase-3含量呈浓度依赖性上升。结论:尿毒症血清促进血管内皮微炎症状态的形成,引起血管内皮细胞氧化损伤,促进血管内皮细胞的凋亡。提示尿毒症患者长期处于慢性炎症状态,心血管疾病的发病风险较高。
Objective:To investigate the in vitro injury effects of sera of rats with uremia on vasular endothelial cells,and explore their clinical significance.Methods:Twenty SD rats were randomly devided into healthy group(sham-operated group) and 5/6 nephrectomy(5/6 NX) for 12 weeks.The chronic renal failure models were established on rats by 5/6 NX.Human umbilical vascular endothelial cells(HUVEC) were treated with sera of rats with uremia and healthy rats of different volume fractions(10%、15%、20%、25%).The cell morphology was observed by immunohistochemical staining;Cell viability was assessed by measuring their ability to metabolize the MTT salt;and Real-Time PCR was employed to detect the expression of IL-6,TNF-α,NF-κB,Caspase-3 mRNA in cells.Results:Compared with healthy serum group,MDA was increased significantly,the expression of IL-6、TNF-α、NF-κB and Caspase-3 mRNA in HUVEC increased in uremic setum group.Conclusion:Treatment with sera of rats with uremia may change the morphology,inhibit the proliferation,promote the apoptosis,enhance the secretion of IL-6、TNF-α、NF-κB and Caspase-3 and lead to the dysfunction of vascular endothelial cells.Patients with uremia are under chromic inflammatory state in a long term,and have a greater risk of developing cardiovasular diseases.
