摘要
目的 研究坛紫菜中酸性多糖的化学结构。方法 采用CTAB结合法、超滤法使酸性多糖PP1与中性多糖PP2分离 ,DEAE -纤维素、琼脂糖凝胶使PP1纯化、紫外光谱、红外光谱、气相色谱、高碘酸氧化、Smith降解等色谱与化学方法测定PP1化学结构。结果 PP1由半乳糖、3- 6 -内醚 -L -半乳糖、甘露糖、葡萄糖组成 ,具α ,β型糖苷键 ,连接方式有 ( 1→ 3)、( 1→ 4)、( 1→ 2 ) 3种、其总糖、3,6 -内醚 -L -半乳糖及硫酸基的含量分别为 85 .41%、5 .95 %、14.0 %。PP1分子量约为 47.9万。
OBJECTIVE To study the chemical structure of the acidic polysaccharide in Porphyra haitanensis. METHODS CTAB and ultrafiltration were used to separate acidic polysaccharide PP1 from neutral polysaccharide PP2. PP1 was purified with DEAEcellulose and agarose jel. The chemical structure of PP1 was detected with the chromatographic and chemical techniques of ultraviolet spectrum, infrared spectrum, gas chromatography, periodic acid oxidation and Smith degradation. RESULTS PP1 consisted of galactose, 36inner etherLgalactose, seminose and glucose. There existed glucosidic bond, and there were three linking ways: (1→3), (1→4) and (1→2). The contents of total saccharide, 36inner etherLgalactose and sulfate radical were 85.41%, 5.95% and 14.0%. The molecular weight of PP1 was 4 790 000. CONCLUSION It is the first time to have elaborate study of the chemical structure of the acidic polysaccharide in Porphyra haitanensis.
