不同温度热疗对人胃癌耐药细胞株SGC7901/DDP的影响

摘要 目的观察热疗对人胃癌耐药细胞株SGC7901/DDP的影响,为临床上应用热疗及热化疗联合治疗耐药胃癌提供理论依据。方法采用四甲基偶氮唑蓝(MTT)法检测不同温度热疗、热化疗对SGC7901/DDP细胞的抑制作用;流式细胞仪及反转录-聚合酶链反应(RT-PCR)分别检测热疗、热化疗对细胞GST-π蛋白及mRNA表达的影响。结果不同温度热疗处理SGC7901/DDP细胞后对顺铂的敏感性均有提高,以43℃时最佳;43℃热疗2h处理前后此细胞的IC50值分别为(11.7±0.6)μg/mL及(7.2±0.7)μg/mL。耐药逆转倍数RF为1.63倍;热疗后尤其是热化疗后SGC7901/DDP细胞GST-π蛋白及mRNA表达明显下降(P<0.01)。结论热疗能提高人胃癌耐药细胞株SGC7901/DDP对顺铂的敏感性,热化疗可能更大限度地抑制SGC7901/DDP细胞增殖;热疗部分逆转其耐药机制可能与下调GST-π蛋白及mRNA表达有关。 Objective To observe the effects of hyperthermia therapy combined with chemotherapy on multi drug resistance in human gastric adenocarcinoma cell line SGC7901/DDP in order to provide theoretical evidence for hyperthermia and thermochemotherapy in clinic. Methods MTT assay was used to detect the growth rate by hyperthermia or thermochemotherapy at different temperature groups (41 ℃ ,42 ℃ ,43 ℃ ,44 ℃ ) ; GST-π protein expression level was analyzed by FCM and GST-π mRNA expression was detected using RT-PCR. Results Trea-ted with different temperature, drug sensitivity of SGC7901/DDP to cisplatin all increased,and the best one is 43 ℃ ;After treatment with hyperthermia at 43 ℃ for 2 h of SGC7901/DDP, the IC50 value decreased from (11.7±0.6)μg/mL to (7.2±0.7)μg/mL, the reversal fold of multiple drug resistance is 1.63;After hyperthermia, the expression of GST-π protein and mRNA was in downward trend, and the thermochemotherapy group is more sig-nificant. Conclusion Hyperthermia can significantly enhance the sensitivity to cisplatin in SGC7901/DDP. Ther- mochemotherapy can inhibit cell proliferation. Hyperthermia had the reversal function on SGC7901/DDP, which may be obtained by the decrease of GST-π protein and mRNA expression level.
