重复加热对肝癌HepG2细胞凋亡的影响 被引量:1

The Effects of Repeated Thermotherapy on the Apoptosis of HepG2 Cells

摘要 目的探讨连续10d加热后残留的HepG2细胞的凋亡率改变及Bc l-2和Bax基因/蛋白的表达变化。方法HepG2细胞经43℃加热,每次80m in,2次/d,共10个循环后,检测其细胞凋亡率、Bc l-2和Bax基因/蛋白的表达。结果热处理前、后HepG2细胞的凋亡率分别为(9.0%±0.8%)、(5.8%±1.3%)(P<0.05);热处理后HepG2细胞的Bax基因/蛋白的表达无明显变化,但Bc l-2基因/蛋白的表达增强,且Bc l-2/Bax比值的增加。结论热处理后HepG2细胞的凋亡率降低与其Bc l-2基因/蛋白的强表达及Bc l-2/Bax比值的增加有关。 Objective To investigate the apoptosis rate and the mRNA and protein expressions of Bcl-2 and Bax in the remained HepG2 cells after successive thermotherapy for 10 days. Methods After the HepG2 cells were treated with ten repeated cycles of heat exposure at 43℃ for 80 minutes twice a day, the rate of apoptosis was analyzed, and the mRNA and protein expressions of Bcl-2 and Bax were detected. Results Before and after treatment with heat, the apoptosis rates of HepG2 cells were (9.0%±0.8% ) and (5.8%±1.3%) respectively(P〈0.05), and there was not significant difference in the mRNA and protein expressions of Bax in HepG2 cells. After the treatment with heat, the mRNA and protein expression of Bcl-2 was increased than that before the treatment, and the ratio of Bcl-2 to Bax was higher. Conclusion After treatment with heat, the apoptosis rate of HepG2 cells was decreased due to the higher expression of Bcl-2 mRNA and protein and higher ratio of Bcl-2 to Bax.
