摘要 目的测定正常兔眼球后注射榄香烯后药物在眼内的分布。方法采用气相色谱法测定正常兔眼单次及连续1周球后注射榄香烯后房水、玻璃体、视网膜和脉络膜中β-榄香烯的浓度。结果单次球后注射榄香烯后1h,房水、玻璃体、视网膜和脉络膜中均检测到β-榄香烯。连续1周注射后,仅视网膜和脉络膜中检测到β-榄香烯,浓度与单次注射相比无明显增加。结论正常兔眼球后注射榄香烯可通过血-视网膜屏障进入眼内,连续用药后无明显蓄积作用。 Objective To determine the distribution of β-elemene in normal rabbit eyes after single and repeated retrobulbar injections. Methods Elemene was injected into the retrobulbar space of normal rabbit eyes at a dose of 5mg. Besides single injection, repeated injections were also performed once a d for one week. Aqueous and vitreous were collected. Retinas and choroids were dissected off and reduced to pulp separately. The concentration of p-elemene in these four compartments were measured by gas chromatography. Results β-elemene was found in aqueous,vitreous,retina and choroid 1 h after single retrobulbar injection. The concentrations of β-elemene in these four compartments decreased in the following order: choroid > retina > vitreous > aqueous. After repeated retrobulbar injections for one week, however, β-elemene was found only in retina and choroid,and the concentration of β-elemene in retina was higher than that in choroid. It was also found that the β- elemene concentration in retina after repeated injections was not significantly different in comparison with that after single injection,whereas β-elemene concentration in choroid decreased markedly. Conclusion Elemene can pass the blood-retinal barrier of normal rabbit eye after retrobulbar injection and does not accumulate significantly after one week of repeated injections, implying that elemene has potential value in ophthalmic therapy.
