摘要
目的:探索简单快速检测榄香烯脂质体磷脂含量的方法。方法:采用氯仿破坏脂质体,硫氰铁铵与磷脂反应生成配位化合物,在468 nm处检测其吸收度,计算其磷脂含量。结果:回归方程为A=10.65C-0.325,在0.10-0.35 mg.mL-1范围内成线性。平均加样回收率为113.3%(n=9),RSD%为7.9%,其RSD值较大,超过5%。日内差异度RSD%为2.1%(n=9),日间差异度RSD%为4.0%(n=9)。榄香烯脂质体的磷脂含量为(19.95±1.76)mg.mL-1,RSD%为8.8%,大于5%。结论:分光光度法测定磷脂含量简单、快速,但尚不够精确,适用于榄香烯磷脂含量的快速粗略分析。
Objective: To detect simply and lastly phospholipid content of elemene liposomes. Methods: Liposomes were destroyed using chloroform, and the coordination compound was generated by reaction of Ammonium Ferrothiocyanate and phospholipid. The absorption was detected at 468 nm, and phospholipid content was calculated. Results: Its regression equation is A=10.65C-0.325, and its linear range was 0.10-0.35 mg.mL^-1. Its average recovery was 113.3% (n=9),RSD% was 7.9% and more than 5%. The RSD of intra-day and inter-day precision was 2.1% (n=9) and 4.0% (n=9). The content of elemene liposomes was (19.95±1.76)mg.mL^-1, RSD% was 8.8% and more than 5%. Conclusion: Spectrophotometric method for determination of phospholipid content was simple and fast, but not accurate enough, so it can be used for detecting phospholipid content of elemene liposomes fastly and roughly.
