摘要
目的了解Fas和Fas配体(FasL)mRNA在兔角膜中的表达情况,探讨其在角膜移植中的作用。方法人工合成特异性寡核苷酸探针,应用原位分子杂交方法检测6只新西兰白兔双眼角膜上皮、角膜基质和角膜内皮层Fas和FasL mRNA的表达。结果正常兔角膜上皮和内皮细胞层有 Fas mRNA表达,基质中无表达;角膜上皮和内皮细胞层有强 FasL mRNA表达,且基质中亦有表达。兔角膜 FasL mRNA表达强于 Fas mRNA的表达。结论角膜表达 Fas和FasL可能与角膜移植存活有关。
Objective To investigate the expression distribution of Fas and Fas ligand(FasL) mRNA on rabbit corneas, and evaluate its effect on corneal allograft survival. Methods Based on the published oligonucleotide sequences of Fas and FasL, two oligonucleotide probes were synthesized. The expression distribution of Fas and FasL mRNA on the corneal epithelium, cornal stroma and corneal endothelium for six New Zealand rabbits were detected with in situ hybridization. Results Fas mRNA expression was present on the normal corneal epithelium and endothelium,and not present on the normal corneal stroma. FasL mRNA was present intensely on the normal corneal epithelium and endothelium,and also present on the corneal stroma. The capacity for corneal expression of FasL mRNA was higher than that of Fas mRNA. Conclusion Fas and FasL expression on cornea may be relative to corneal allograft survival.
