玫参胶囊对去卵巢骨质疏松大鼠骨密度及骨代谢的影响 被引量:5

The effects of Meishen Capsule on bone mineral density and bone metabolism of osteoporosis rats induced by ovariectomy

摘要 目的:研究玫参胶囊对骨质疏松大鼠骨密度及骨代谢的影响。方法:SD雌性大鼠60只随机分为6组:假手术组、模型对照组、更年欣胶囊组和玫参胶囊高、中、低剂量组。采用摘除双侧卵巢法建立骨质疏松模型,术后10 d开始给药,连续给药90d,检测大鼠骨密度(BMD)、骨矿含量(BMC)、血清钙(Ca)、血清磷(P)、血清骨钙素(BGP)、血清碱性磷酸酶(ALP)和血清抗酒石酸酸性磷酸酶(TRAP)含量。结果:(1)去卵巢大鼠BMD含量变化:与模型对照组比较,玫参胶囊6.4g/kg剂量组腰椎及右股骨BMD均有显著的差异,玫参胶囊12.8g/kg剂量组全身、腰椎及右股骨BMD均有显著的差异。(2)去卵巢大鼠右股骨BMC含量变化:与模型对照组比较,玫参胶囊6.4g/kg剂量组有显著的差异,玫参胶囊12.8g/kg剂量组有非常显著的差异。(3)去卵巢大鼠血清Ca含量变化:与模型对照组比较,玫参胶囊12.8g/kg剂量组有显著的差异。(4)去卵巢大鼠血清P含量变化:与模型对照组比较,玫参胶囊12.8g/kg剂量组有非常显著的差异。(5)去卵巢大鼠血清BGP含量变化:与模型对照组比较,玫参胶囊6.4g/kg及12.8g/kg剂量组均有非常显著的差异。(6)去卵巢大鼠血清ALP含量变化:与模型对照组比较,玫参胶囊6.4g/kg剂量组有显著的差异,玫参胶囊12.8g/kg剂量组有非常显著的差异。(7)去卵巢大鼠血清TRAP含量变化:与模型对照组比较,玫参胶囊3.2g/kg剂量组有显著的差异,玫参胶囊6.4g/kg及12.8g/kg剂量组均有非常显著的差异。以上实验结果中,玫参胶囊有效剂量在6.4~12.8g/kg之间,但以12.8g/kg效果尤著。结论:玫参胶囊6.4g/kg和12.8g/kg剂量组能有效升高去卵巢骨质疏松大鼠BMD与BMC含量,降低血清中Ca、P、BGP、ALP及TRAP含量,对去卵巢所致大鼠骨质疏松有一定的治疗作用。 Objective: To study the effects of Meishen capsule on bone mineral density and bone Metabolism in osteoporosis rats induced by ovariectomy.Methods: 60 SD female rats were randomly divided into 6 groups,which were sham-operated group,osteoporosis model group,Gengnianxin capsule group,Meishen capsule with high dose,medium dose and low dose groups.Osteoporosis model was established by ovariectomy.The rats were treated consecutively for 90 days with Meishen capsule after 10 days of ovariectomy.The effects of Meishen capsule on Bone mineral density(BMD),Bone mineral content(BMC),Calcium(Ca),Phosphorus(P),Bone gamma-carboxyglutamic-acid-containing proteins(BGP),Alkaline phosphatase(ALP)and Tartrate-resistant acid phosphatase(TRAP).Results:(1) The variation of bone mineral density in ovariectmoized rats: compared with model group,the Meishen capsule group(6.4g/kg) has the significant difference in bone mineral density in lumbar vertebrae and right femur(P0.05),Meishen capsule group(12.8g/kg) has the significant difference in bone mineral density in whole body 、lumbar vertebrae and right femur(P0.05 or P 0.01).(2) The variation of bone mineral content in the right femur in ovariectmoized rats: compared with model group,the Meishen capsule group(6.4g/Kg) has the significant difference in the variation(P0.05) and the Meishen capsule group(12.8g/kg) has the extremely significant difference in data(P0.01).(3)The variation of the content of Calcium in ovariectmoized rats: the Meishen capsule(12.8g/kg) has the significance difference compared with model group(P0.05).(4) The variation of the content of Phosphorus in ovariectmoized rats:the Meishen capsule(12.8g/kg) has the extremely significance difference compared with model group(p0.01).(5) The variation of the content of bone gamma-carboxyglutamic-acid-containing proteins in ovariectmoized rats: the Meishen capsule(6.4g/kg and 12.8g/kg) has the extremely significance difference compared with model group(P0.01).(6) The variation of the content of alkaline phosphatase in ovariectmoized rats: compared with model group,the Meishen capsule(6.4g/kg) has the significance difference(P0.05) and the Meishen capsule(12.8g/kg) has the extremely significance difference(P0.01).(7) The variation of the content of tartrate-resistant acid phosphatase in serum of the ovariectmoized rats: the Meishen capsule(3.2g/kg) has the significance difference compared with model group(P0.05) and the Meishen capsule(6.4g/kg and 12.8g/kg) has the extremely significance difference in the results(P0.01).All the results below demonstrated that the effective dose of Meishen capsule is between 6.4g/kg and 12.8g/kg,and the dose of 12.8g/kg has the significant effective result.Conclusion: The Meishen capsule group(6.4g/kg and 12.8g/kg) have the effect to increase the content of BMD and BMC,also can decrease the content of Ca,P,BGP,ALP and TRAP in serum in ovariectmoized rats.The Meishen capsule seemed had the effect in osteoporosis in ovariectmoized rats.
