摘要
目的研究D-半乳糖致衰老大鼠海马及下丘laminA/C、prelaminA、FACEl的水平变化,并进-步明确石杉碱甲对D-半乳糖诱导的这些蛋白变化的影响及其意义。方法36只健康雄性SD大鼠随机分成对照组、D-半乳糖治疗组和D-半乳糖联合石杉碱甲组。D-半乳糖组每天皮下注射D-半乳糖300mg/Kg,共8W;D-半乳糖联合石杉碱甲组每天D-半乳糖300mg/Kg联合石杉碱甲0.1mg/Kg共同皮下注射,共8W。对照组皮下注射等量生理盐水。应用蛋白免疫印迹杂交(Western blotting)方法检测3组大鼠海马、下丘laminA/C、prelaminA和FACEl的表达。结果海马中,laminA在对照组、D半乳糖+石杉碱甲组、D-半乳糖组中逐渐降低,laminC水平逐渐升高,prelaminA在对照组中最高,D-半乳糖组与D-半乳糖+石杉碱甲组无明显差异;FACEl在D-半乳糖+石杉碱甲组水平最高,对照组中水平最低。下丘中,laminA水平在D-半乳糖+石杉碱甲组最高,D-半乳糖组中最低;D-半乳糖组和D-半乳糖+石杉碱甲组laminC水平较对照组升高,D-半乳糖组与D-半乳糖+石杉碱甲组之间的差异无统计学意义;prelaminA水平在对照组中最低,D-半乳糖+石杉碱甲组中最高;FACE1水平在对照组、D-半乳糖+石杉碱甲组、D-半乳糖组中逐渐升高。结论LaminA降低、laminC及FACE1升高预示着D-半乳糖诱导的海马及下丘衰老。石杉碱甲可以在-定程度上改善部分指标的变化,说明它有-定的延缓海马及下丘衰老的作用。
Objective To investigate the changes of lamin A/C, prelamin A, FACE1 in the hippocampus and inferior colliculus of D-galactose treated aging rats. And also, to identify the effects and significance of huperzine A on the proteins changes induced by D-galactose. Method Thirty-six healthy male SD rats were randomly assigned into three groups: normal group, D-galactose treatment group and D-galactose combined with huperzine A treatment group. D-galactose group were injected subcutaneously with D-galactose 300mg/Kg/d for eight weeks. D-galactose combined with huperzine A treatment group were injected subcutaneously with D-galactose 300mg/Kg/d and huperzine A 0. 1mg/Kg/d for eight weeks. The nor- mal group were injected with equal amount of normal saline every day. Then Western blotting was used to measure the ex-pression of lamin A/C, prelamin A and FACE1 in the hippocampus and inferior colliculus. Results In the hippocampus, lamin A were gradually decreased in normal, D-galactose combined with huperzine A and D-galactose treated groups. While, lamin C was increased in the three groups gradually. Prelamin A was highest in the normal group and there was no significant difference between D-galactose combined with huperzine A and D-galactose treated groups. FACE1 was highest in the D- galactose combined with huperzine A group and lowest in the nornal group. In the inferior colliculus, lamin A was highest in the D-galactose combined with huperzine A treated group and lowest in the D-galactose treated group. The levels of lamin C in D-galactose treated group and D-galactose combined with huperzine A treated group have no significant difference, but they were both higher than normal group. As for prelamin A, it was highest in the D-galactose combined with huperzine A group and lowest in the normal group. The levels of FACElin normal, D-galactose combined with huperzine A and D- galactose treated group were increased gradually. Conclusions Lower levels of lamin A~ higher levels of lamin C and FACE1 indicate hippocampus and inferior colliculus aging in D-galactose treated rats. Huperzine A can improve some of the proteins changes indicating its anti-aging role in the hip-pocampus and inferior colliculus.
