咬合创伤大鼠颞下颌关节囊神经中蛋白基因产物9.5的表达变化 被引量:3

摘要 目的:研究咬合创伤大鼠颞下颌关节囊神经中蛋白基因产物9.5(PGP9.5)的表达变化。方法:在大鼠右上第一磨牙面粘接正畸方丝,使咬合抬高约0.5mm,形成右侧磨牙早接触的创伤性咬合。分别于建模后1,3,7,14,28d进行免疫组化染色和实时荧光定量PCR检测,测定各组大鼠颞下颌关节囊神经PGP9.5蛋白和mRNA的表达。结果:1d组关节囊神经PGP9.5蛋白表达量较对照组降低,无统计学意义,3d组明显升高(P<0.05),7d达到峰值(P<0.05),14d、28d逐渐降低。1d组关节囊神经PGP9.5mRNA相对表达量较对照组升高(P<0.05),3d达到峰值(P<0.05),7d、14d、28d逐渐降低。结论:咬合创伤早期关节囊神经PGP9.5蛋白和mRNA的表达量升高。 Objective: To investigate the effect of traumatic occlusion on the expression of Protein Gene Product 9.5 (PGP9.5) in the nerves of temporomandibular joint (TMJ) capsules of rats. Methods: The occlusal surface of the right first maxillary molar was raised 0.5ram by bonding an orthodontic square wire on its surface, to establish traumatic occlusion an- imal models. The TMJs were prepared for immunohistochemical staining, and TMJ capsules were prepared for real-time PCR analysis at 1, 3, 7, 14 and 28 days after teeth operation. Result: Expression of PGP 9.5 decreased slightly on the 1st day (P〉0.05), increased significantly on the 3th day (P〈0.05), reached its peak on the 7th day (P〈0.05), and decreased on the 14th and 28th day. Expression of PGP 9.5 mRNA increased slightly on the 1st day (P〈0.05), reached its peak on the 3th day (P〈0.05), and decreased on the 7th, 14th and 28th day gradually. Conclusion: Traumatic occlusion was found to in- crease the expression of PGP 9.5 and PGP 9.5 mRNA in the early stage of traumatic occlusion.
