糖尿病心肌梗死大鼠TGF-β1和CTGF表达变化及HGF的干预作用 被引量:6

摘要 目的:探讨糖尿病心肌梗死大鼠心肌TGF-β1和CTGF表达的变化以及肝细胞生长因子(HGF)对其影响。方法:6周龄Wistar大鼠90只随机分为5组:对照纽、糖尿病假手术纽、心肌梗死组、糖尿病心梗组和HGF治疗组,每组各18只。以高糖高脂饲料+腹腔注射链尿佐菌素(STZ)诱导建立2型糖尿病大鼠模型后再建立急性心肌梗死(AMI)模型。糖尿病模型大鼠成模后次日,HGF治疗组每天皮下注射HGF,分别观察4周及8周,检测各组心肌组织TGF-β1、CTGF的mRNA及蛋白表达情况。结果:对照组大鼠的TGF-β1、CTGF mRNA及蛋白水平表达微弱;糖尿病心梗组4周后TGF-β1、CTGF mRNA及蛋白水平表达明显升高(P<0.05),8周后表达进一步升高(P<0.01);给予HGF治疗4周后,可以部分降低TGF-β1、CTGFmRNA及蛋白水平的表达(P<0.05);给予HGF治疗8周后,可以明显降低TGF-β1、CTGF mRNA及蛋白水平的表达(P<0.01)。结论:糖尿病心肌梗死大鼠心肌TGF-β1、CTGF的表达明显增高,HGF治疗后能够降低TGF-β1、CTGF的表达,对糖尿病心肌梗死后心肌纤维化的发生有抑制作用。 To determine the changes in and the effects of hepatocyte growth factor (HGF) on transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CTGF) expression in rats with streptozotocin-induced diabetes mellitus (DM) and concomitant acute myocardial infarction (AMI). Methods: Ninety Wistar rats 6 weeks of age were randomly assigned to control group ( n = 18) , group sham-operated DM (n=18), group AMI (n= 18), group DM-AMI (n = 18) and group HGF (n =18), respectively. The rats were fed with sugar- and lipid-rich foods supplemented with streptozotocin (STZ) peritoneal injection for construction of type 2 DM model followed by AMI model. Group HGF received subeutaneous HGF injection on day 2 following construction of DM model. This entailed measurement of the expression of TGF-β1 and CTGF mRNA and protein at weeks 4 and 8. Results: The TGF-β1 and CTGF mRNA and protein expression was barely detectable in control group (both P 〉 0. 05) and significantly increased at week 4 ( both P 〈 0.05 ) and progressively augmented at week 8 (both P 〈0.01 ) in group DM-AMI. Treatment with HGF for 4 weeks led to partly suppressed TGF-β1 and CTGF InRNA and protein expression ( both P 〈 0. 05 ) , which was further attenuated at week 8 (both P 〈 0.01 ). Conclusion: The augmented TGF--β1 and CTGF expression in rats with streptozotoein-indueed DM and concomitant AMI could 13e attenuated by the HGF therapy, suggesting that HGF may inhibit tile progression of post-infhrction myoeardial fibrosis in rats with DM.
