摘要
目的:探讨转化生长因子β(1TGF-β1)在自发性高血压大鼠(SHR)左心室肥厚发生过程中的作用和咪哒普利、厄贝沙坦在抑制TGF-β1表达方面的作用效果。方法:选用13周龄SHR30只,随机分为SHR组、厄贝沙坦组、咪哒普利组;另选同源同系WKY大鼠10只,作为正常对照组(WKY组)。应用大鼠尾压心率测定仪测量血压,应用SABC免疫组化及RT-PCR检测左心室肌TGF-β1蛋白及mRNA水平。结果:血压、左心室肌TGF-β1蛋白和mRNA的表达SHR组明显增加;咪哒普利组、厄贝沙坦组降低;咪哒普利组的表达低于厄贝沙坦组。结论:SHR的左心室肌TGF-β1蛋白和mRNA的表达明显增高;咪哒普利、厄贝沙坦不仅能良好地控制血压而且可以抑制SHR左心室肌TGF-β1蛋白和mRNA的表达;咪哒普利对TGF-β1蛋白和mRNA表达的抑制作用强于厄贝沙坦。
Objective:To explore the role of transforming growth factor-β1 (TGF-β1) in the development of left ventricular hypertrophy in spontaneously hypertensive rat (SHR) and the effects of imidapril and irbesartan on TGF-β1 expression. Methods:Thirty 13-week-old SHRs were randomly divided into SHR group,imidapril group,and irbesartan group,and 10 homogenous Wistar-Kyoto (WKY) rats were selected as normal control group. The blood pressure of rats was monitored. The levels of TGF-β1 protein and mRNA were determined by immunohistochemical SABC method and reverse transcription polymerase chain reaction, respectively. Results:The blood pressure and the levels of TGF-β1 protein and mRNA significantly increased in SHR group and decreased in imidapril and irbesartan groups. The levels of TGF-β1 protein and mRNA in imidapril group were lower than those in irbesartan group. Conclusion:imidapril and irbesartan could not only control blood pressure but also inhibit the expressions of TGF-β1 protein and mRNA in the left ventricle myocardium of SHR. Such effects are stronger in imidapril group than in irbesartan group.
