摘要
为了得到更利于细胞融合的原生质体 ,研究了几个影响金针菇担孢子原生质体制备的最重要因子 ,当用 0 .6mol/L Mg SO4 .7H2 O作渗透压稳定剂 ,在 35℃、2 %溶壁酶中酶解2 .5h后 ,可获得纯净且量多的原生质体 ,其产量最高达 6× 1 0 7个 /m L,原生质体大小比较均一 ,直径约 8.2
Several important factors affecting formation of Protoplasts of Flammulina velutipeswere successfully reported in order to obtain the optimal protoplasts of cell fu- sion.The optimal conditions for protoplast formation:using 0 .6mol/L Mg SO4.7H2 O as the osmotic stabilizer,Flammulina velutipeswere treated with2 % lywallzyme at3 5℃ , numorous and pure protoplasts could be obtained after 2 .5hours.The highest yield of protoplasts amounted to 6× 1 0 7per m L.
