microRNA-10b对人食管癌细胞系EC9706迁移和侵袭的影响 被引量:3

摘要 目的:探讨微小RNA-10b(microRNA-10b,miR-10b)对食管癌细胞系EC9706迁移和侵袭能力的影响。方法:构建pEGFP-miR-10b真核表达载体,在食管癌细胞系EC9706中稳定表达miR-10b,利用real-time PCR检测其表达水平,应用划痕试验,Transwell小室实验分析迁移和侵袭能力的改变。结果:转染miR-10b载体后,real-time PCR检测其表达水平显著高于对照组(P<0.05),miR-10b的表达能够显著促进EC9706细胞的迁移和侵袭能力(P<0.05)。结论:miR-10b能够促进食管癌细胞系EC9706的迁移和侵袭,可作为评价食管癌转移能力的指标。 Objective:To explore the effect of miR-10b on the metastasis and invasion of human esophageal cancer cell line EC9706.Methods:The eukaryotic expression vector of pEGFP-miR-10b was constructed and stably transfected into EC9706 cells.The expression level of miR-10b was determined by real-time PCR.The migration of EC9706 cells transferred with miR-10b was accessed by Scratch assay.Meanwhile,the migration and invasion capacities of EC9706 cells were accessed by Transwell assay.Results:The expression level of miR-10b on EC9706 cells transferred with miR-10b significantly increased compared with that in control group(P0.05).The migration and invasion capacity of EC9706 cells transferred with miR-10b markedly increased(P0.05).Conclusion:miR-10b could increase the migration and invasion capacities of EC9706 cells,suggesting that miR-10b might be a novel value predicting marker for the metastasis capacity of human esophageal cancer.
