摘要
目的:优选鹿衔草中总黄酮的酶联半仿生法提取工艺。方法:以总黄酮提取率为指标,在单因素试验基础上,选取加酶量、料液比、酶解时间、提取时间、提取温度5个因素进行Box-Behnken中心组合设计,利用响应面法优选提取工艺。结果:优选的提取工艺为加酶量1.25 mg.g-1,料液比1∶65,酶解时间1.0 h,提取时间1.75 h,提取温度85℃,总黄酮提取率达5.54%,与预测值5.68%较为接近。结论:采用响应面法优选的提取工艺合理可行,具有良好预测性。
Objective: To optimize enzyme-linked semi-bionic extraction technology of total flavanoids from Pyrola decorata.Method: With yield of total flavanoids as index,based on single factor test,Box-Behnken central composite design was adopted by selecting the amount of enzyme,material-liquid ratio,enzymolysis time,extraction time and temperature as factors,extraction technology was optimized by response surface methodology.Result: Optimum extraction conditions were as follows: the amount of enzyme 1.25 mg.g-1,ratio of material to liquid 1 ∶ 65,enzymolysis time 1.0 h,extraction time 1.75 h,extraction temperature 85 ℃.Under this conditions,yield of total flavanoids was up to 5.54%,which was close to the predicted value(5.68%).Conclusion: Optimized technology was reasonable and feasible with good predictability.
