三株分泌抗细粒棘球蚴囊液抗原单克隆抗体的杂交瘤细胞的建立和初步鉴定 被引量:6

摘要 以羊肝细粒棘球蚴囊液抗原免疫BaLb/c鼠脾细胞与SP2/o骨髓瘤细胞常规方法融合。用人肝棘球蚴囊液抗原间接ELISA法检测,获得3株稳定分泌抗体的杂交瘤细胞株,2G_4、2F_1和2F_(10)。对人肝包囊液抗原ELISA的终点分别为1∶204800,1∶819200和1∶3200。对纯化羊肝包囊液抗原致敏羊血球IHA的终点均为1∶512,但3株单抗等量混合后IHA滴度达1∶4096。对猪囊尾蚴抗原的ELISA滴度分别为1∶4000、1∶2000和1∶2000。对泡球蚴抗原ELISA滴度分别为1∶2000、1∶1000和1∶2000。交叉阻断ELISA试验的结果初步显示此3株单抗可能识别不同的抗原表位。3株细胞在液氮中冻存两年复苏后,仍继续稳定分泌抗体。 Three hybridoma cell lines (2G4, 2F1 and 2F10) constantly secreting antibodies against sheep hydatid cyst fluid antigen were obtained using routine technique by fusion of immunized Balb / c mice spleen cells and myaloma SP 2 / o cells. The antibody detection method used to screening hybrids was the indirect ELISA with human liver hydatid cyst fluid as antigen. The antibody titre of ascites fluid of 2G4 2F1 and 2F10 in ELISA were 1 : 204800, 1 : 819200 and 1 : 3200 respectively. The IHA titre of all these McAbs was 1 : 512, however, in pooled ascites fluid IHA titre increased to 1 : 4096. Results of cross blocking ELISA suggested that these McAbs might be capable of recognizing different antigenic epitopes.
