Comparison of in vitro maturation of swine oocyte in different months

摘要 【目的】分析卵母细胞发育与不同季节(月份)的相关性,为提高猪卵母细胞的体外成熟效率奠定基础。【方法】2012年2~6月从南宁市某屠宰场收集猪卵巢采集卵母细胞,经体外成熟(IVM)40-44h,然后随机挑出成熟后的卯母细胞进行地衣红染色,相差显微镜下观察、记录卵母细胞发育成熟的各个时期,并分析卵母细胞发育时期与不同月份温度、湿度的相关性。【结果】随着月份的增加,GV期和GVBD期卵母细胞所占比例整体上呈下降趋势,AT期所占比例呈上升趋势,除5月外,MII期所占比例整体上也呈上升趋势,6月MII期所占比例最高,为42.31%。2、3、4月的GVBD期所占比例显著高于6月所占比例(4.40%)(P〈0.05)。从卵母细胞发育时期与不同月份温度、湿度的相关性分析结果可知,GV期、GVBD期、MI期3个时期与温度变化呈负相关,与湿度变化呈正相关;而AT期、MII期与温度变化呈正相关,与湿度变化呈负相关。【结论】不同月份猪卵母细胞的ⅣM效果存在一定差异。在高湿低温月份,GV期和GVBD期的卵母细胞所占比例较大,MII期所占比例较小;而在低湿高温月份,Gv期和GVBD所占比例较小,MII期所占比例较大。 [Objective]To provide references for improving in vitro maturation efficiency of swine oocyte, the correlation between oocyte development and different seasons (months) was analyzed. [Method]From February to June 2012, swine oocytes were collected from a slaughterhouse at Nanning. After 40 to 44 h in vitro maturation (IVM), the mature oocytes were picked out for orcein staining, observed and recorded. The correlation between oocyte development stage and tempera- ture/humidity in different months was assayed. [ Result ] With the increase of months, the proportion of oocytes at GV stage and GVBD stage showed a downward trend overall, and AT stage was rising. Except May, the proportion of ooeytes at MII stage also showed an ascendant trend. The proportion of oocytes at MII stage in June was the highest(42.31%). The proportion of GVBD period in February and April were significantly higher than that in June (4.40%)( P〈0.05 ). From the correlation analysis results, GV, GVBD and MI were negatively correlated with temperature changes, and positively corre- lated with humidity changes, while AT stage and MII stage were positively correlated with temperature changes, and nega- tively correlated with humidity change. [Conclusion]IVM effect of oocytes varied in different months. In cold month with high humidity, the proportion of oocytes at GV stage and GVBD stage were larger, and the proportion of oocytes at MII stage was smaller, while in hot month with low humidity, the proportion of oocytes at GV stage and GVBD stage were smaller, and the proportion of oocytes at MII stage was larger.
