摘要
从屠宰场收集卵巢,用注射器从卵泡中吸出卵丘-卵母细胞复合体,对猪卵母细胞体外成熟的条件进行研究,根据卵母细胞体外成熟时的核成熟率、胞质成熟及体外受精时精子的穿卵速度,合适的体外成熟培养时间为42~45h.卵母细胞周围卵丘细胞的多少及其排列紧密状态与卵母细胞的体外成熟率密切相关。卵泡大小与卵母细胞的体外成熟率有关,大卵泡卵优于小卵泡卵。PMSG促进猪卵母细胞体外成熟。
Porcine ovarian oocytes were used in the studies on influencing factors of in vitro maturation(IVM). It was showed that the oocytes must be culturdin vitro for 42~45 hrs before they canbe fertilized as indicated by the nuclear maturation rates,ultrastructural changes and the time forsperm penetration. There was a good correlation beitween the number and arrangement of thecumulus cells and the IVM rates of the oocytes. The IVM rate of class A oocytes with 5 or morelayers of cumulus cells was 73.5%(61/83) , the IVM rate for class B oocytes with less than 5layers of cumulus cells was 54.3%(51/94),and that for class C oocytes with no cumulus at allwas 29.4%(12/62). The IVM rate of the oocytes with compact cumulus mass was 60.5%(26/43) , and that of the oocytes with loose cumulus or no cumulus cells was 9.1%(1/11). Thefollicular size had effect on the IVM rates of oocytes also. The IVM rates of oocytes in the largefollieles(4~8mm in diameter)and the small follicles(2~3mm in diameter)were 66.7%(22/33)and 21.7%(10/46) respectively. The proportion of oocytes reaching metaphase II after 42 hrsculture was significantly increased by the addition of PMSG from 17.8%to 67.7%. A hishcumulus expansion rate(92.7%)was found in the oocytes cultured in the medium with PMSG,while those cultured without PMSG showed no expansion.
