摘要
目的研究生长激素(GH)和生长激素受体(GHR)在胃癌患者中的表达及临床意义。方法收集84例胃癌患者术前血清、术后癌组织标本分别作为血清实验组和组织实验组,20例健康体检者血清作为血清对照组,84例胃癌患者术后相应癌旁正常组织作为组织对照组,分别采用酶联免疫吸附法和免疫组化法检测血清GH与组织GHR的表达情况。结果①血清实验组GH平均水平及阳性率与血清对照组相比差异有统计学意义(P<0.05),血清GH水平与肿瘤大小、淋巴结转移密切相关(P<0.05);②组织实验组GHR阳性率与组织对照组相比差异有统计学意义(P<0.05),组织GHR水平与胃癌分化程度密切相关(P<0.05);③血清GH与组织GHR的表达呈负相关(r=-0.405,P=0.001)。结论血清GH和组织GHR表达水平与胃癌的发生发展密切相关,可为胃癌的诊断、预后评估及内分泌治疗提供理论基础。
Objective To investigate the expression and clinical significance of growth hormone （GH） and growth hormone receptor（GHR） in patients with gastric carcinoma. Methods The preoperative sera and postoperative tumorous tissues of 84 patients with gastric carcinoma served as seral and tissular experimental group, respectively, the sera of 20 normal persons as seral control group, and the postoperative corresponding para-tumorous tissues of 84 patients with gastric carcinoma as tissular control group, enzyme-linked immunosorbent assay（ELISA） and immunohistochemistry （IHC） methods were respectively used to detect the expression of serum GH and tissue GHR. Results①The differences of the average levels and positive rate of GH between seral experimental and control groups were statistically significant （P 〈 0.05 ）. Serum GH levels was closely correlated to tumor size and adenoid metastasis （P 〈 0.05 ） ; ② The differences of the positive rate of tissue GHR between tissular experimental and control groups were statistically significant （ P 〈 0.05 ）. Tissue GHR levels was highly correlated to differentiation of tumor（P 〈 0. 05 ）. ③ Serum GH levels was negatively correlated to tissue GHR （ r = - 0. 405, P = 0. 001 ）. Conclusion The expression levels of serum GH and tissue GHR are significantly correlated with tumorigenesis and development of gastric carcinoma, which may provide a potential use for the diagnosis, prognosis evaluation and endocrine therapy of the patients.
