P_5增强卵巢癌细胞顺铂敏感性的体外研究

摘要 目的:探讨凋亡诱导核苷P5增强卵巢癌细胞顺铂(DDP)敏感性的作用,为临床改善卵巢癌患者的预后提供新方法。方法:选择卵巢癌细胞株COC1,以MTT法、Hoechst染色法,观察P5对卵巢癌细胞的生长抑制作用及其对DDP化疗的增敏作用。结果:小剂量P(525μg/mL)和大剂量P(5200μg/mL)分别联合DDP共同作用于COC1细胞,DDP IC50值由2.51μg/mL降至1.56~1.52μg/mL。Hoechst染色法观察,小剂量P5和大剂量P5联合DDP作用于COC1细胞株,细胞凋亡明显增加。结论:小剂量P5与大剂量P5均能增加COC1细胞对DDP的敏感性,P5DDP化疗增敏作用与其增加肿瘤细胞凋亡有关。 Objective: To investigate the effect of P5 induced by apoptosis in sensitivity of cisplatin(DDP) on ovarian carcinoma cells and provide new way for improving ovarian cancer patients'prognosis in clinical. Methods:Selecting ovarian cancer strain COC 1, MTT assay and Hoechst fluorescence staining were used to observe the sensitization effect to cisplatin chemotherapy and the inhibition of P5 on growth of ovarian cancer cells. Results: Low-dose (25μg/mL)P5 and high-dose (200 μg/mL)P5 with different concentrations of DDP were added to COC 1 cells respectively, the DDP' s IC50 reduced from 2.51 p.g/mL to 1.56-1.52 μg/mL. By Hoechst fluorescence staining, apoptosis increased obviously when low-dose P5 and high-dose P5 combined with DDP were added to COC 1. Conclusion: Both of the low-dose P5 and high-dose P5 could increase the sensitivity of cisplatin in ovarian carcinoma COCI cells, the effect of P5 increasing the sensitivity of cisplatin is related to increasing apoptosis of cancer cells.
