高效液相色谱法测定鱼鳞中的Ⅰ型胶原蛋白的含量 被引量:5

摘要 目的建立反相高效液相色谱法测定鱼鳞中Ⅰ型胶原蛋白含量的方法。方法以ZORBAX 300SB-C18色谱柱(4.6mm×250mm,5μm)为分离柱,以(A)5%乙腈+0.05%三氟乙酸(TFA)和(B)90%乙腈为流动相,线性梯度洗脱20min,流速1.0mL.min-1,紫外检测波长230nm。结果Ⅰ型胶原蛋白在56~4681μg.mL-1浓度范围内与峰面积具有良好的线性关系(r=0.9997),精密度,RSD 0.83%(n=6),加标平均回收率为100.38%,RSD1.87%。结论该方法简便快速、灵敏、准确,为鱼鳞Ⅰ型胶原蛋白的质控与定量检测提供了1种新的测定方法。 Objective A simple and sensitive high performance liquid chromatography method was devel- oped for the determination of type I collagen in fish scale. Methods A good chromatographic analysis of the type I collagen was achieved using a ZORBAX 300SB-C18 column (4. 6mm×250mm, 5μm) and the mobile phase contained two solvents: (A) 5% acetonitrile with 0. 05% trifluoroacetic acid (TFA) and (B) 90% acetonitrile with a linear gradient program of A-B(v/v) for 20 minutes. The flow rate was 1. 0 mL · min-land the detection wavelength was set at 230nm. Results It existed a good linear relation- ship between the concentration of the type I collagen sample and the chromatographic peak area ran- ging from 56~4681μg · mL 1, with a 0. 9997 correlation coefficient. Precision, the relative standard deviation o{ six parallel injections was 0.83%. The average recovery was 100. 38%, RSD1.87%. Con- clusion This validated method is very convenient, easy to apply, highly sensitive and precise. It pro- vides a new determination method for quality control and quantitative detection of type I collagen in fish scale.
