摘要
1–乙基–3–甲基溴代咪唑([Emim]Br)离子液体对牛骨胶原具有一定的溶解作用,溶解后以水为沉淀剂得到再生物质.对再生物质进行FTIR、UV、SDS-PAGE凝胶电泳,SEM和DSC分析以确定再生物质成分和结构,从而确定牛骨胶原在离子液体中的变性过程.结果表明:再生物质不存在胶原特有的三股螺旋结构,但仍存在肽键结构,为小分子质量多肽,说明溶解过程中离子液体使牛骨胶原变性,不仅对牛骨胶原的网状结构有破坏作用,同时使牛骨胶原三股螺旋结构和肽链遭到破坏,胶原发生不可逆变性.
1-ethyl-3-methylimidazolium bromide([Emim]Br)ionic liquid was used to resolve bovine bone collagen. Deionized water was then employed as precipitant to get the regenerated substance.Fourier transform infrared spectroscopy(FTIR),ultraviolet spectroscopy(UV),SDS-Polyacrylamide gel electrophoresis(SDS-PAGE),scanning electron microscope(SEM)and differential scanning calorimetry(DSC)have been used to identify the chemical components and structure of the regenerated substance,which can indicate the denaturation of bovine bone collagen in ionic liquids.The results indicated that the regenerated substance turns out to be small molecular polypeptide without helical structure,but still has peptide bonds.The crystalline portion of the bovine bone collagen was destroyed and the collagen denaturation is irreversible,revealing the ionic liquid could lead to the denaturation of bovine bone.
