摘要
目的:总结马钱子碱的抗肿瘤作用及其相关抗肿瘤机制。方法:应用PuMed及CNKI期刊全文数据库,以"马钱子碱和抗肿瘤作用"为关键词,检索1992-01-2012-01相关文献,共收集文献126篇。纳入标准:1)马钱子碱的抗肿瘤作用;2)马钱子碱的抗肿瘤作用机制。根据纳入标准,符合分析的文献30篇。结果:无论是体内还是体外,马钱子碱都有显著的抗肿瘤作用。目前研究发现,马钱子碱的抗瘤谱主要包括一些肝癌细胞系,如SMMC 7221、Heps和H22;乳腺癌细胞系,如MDA-MB-231和MCF-7;血液系统肿瘤细胞系,如K562和U266等。马钱子碱的抗肿瘤机制包括诱导肿瘤细胞凋亡、下调一些促肿瘤生长和转移细胞因子的表达,同时上调一些抑制肿瘤生长和转移细胞因子的表达,抗血管生成,逆转肿瘤细胞的多药耐药性。结论:无论是体内试验还是体外实验,马钱子碱对多种肿瘤细胞都有显著的抗肿瘤作用并且涉及到多种抗肿瘤作用机制。
OBJECTIVE: To review advances of the anti-tumor effect and the involved mechanisms of brucine. METHODS: The related literature of anti-tumor effect of brucine was retrieved in PubMed and cnki from 1992 01-2012-01. Key words were brucine and anti-tumor effect, and 126 articals were obtained. The included criteria:l) Anti-tumor effect of brucine. 2) Anti-tumor mechanism of of brucine. Thirty documents were reviewed according to the inclusion criteria. RE- SULTS: Brucine has antitumor effect in vitro and in vivo. The current studies found that brucine has growth inhibitory effects on hepatoma cell line SMMC 7221, Heps and H22. Moreover, brucine can inhibit human breast cell line MDA-MB-231 and MCF-?,human chronic myeloid leukemia cell line K562 cells and multiple myeloma (MM) cell line U266,etc. The anti-tumor mechanism of brucine involves apoptosis, anti-angiogenesis, decreasing VEGF and TNF-aex- pression,increasing IL-12 expression and reversal of tumor ceil multidrug resistance. CONCLUSION: Brucine has antitu- mor effect on many kinds of tumor cells both in vitro and in vivo by various mechanisms.
