摘要
为筛选组织培养特性优良的大麦品种并建立高效的再生体系,选用西北地区主栽品种甘啤3号、甘啤4号、甘啤6号及德国引进品种玛俐、博乐共5个大麦品种,分别研究了基因型、培养基成分、种子处理方式等因素对大麦成熟胚组织培养的影响。结果表明,不同大麦品种的出愈率、绿点率和绿苗率均存在极显著差异,其中博乐的综合效应最好。脱分化培养基A3、分化培养基C2是适合进行再生体系构建的培养基。胚刮碎与纵切处理的愈伤组织诱导率显著高于横切。甘啤3号、甘啤4号、博乐的愈伤组织诱导频率和分化绿苗率均较高,是适合进行遗传转化的受体材料。
In order to optimize a regeneration system of tissue culture from mature embryos in barley and select ideal genotypes for transgenic research,five barley varieties(Ganpi 3,Ganpi 4,Ganpi 6,Marine and Belana) were selected to determine the influence of genotypes,media and different seed cutting on the embryo derived callus induction and plant regeneration.The results indicated that the callus induction rate,regeneration rate of mature embryo was significant difference in 5 barley varieties.Callus induction rate of Belana is higher than others.Media A3 and C2 were suitable to the plant regeneration.The callus induction rate of seed scraping broken and seed cutting longitudinally was higher than that of cutting laterally.Among the five barley cultivars Ganpi 3,Ganpi 4 and Belana were identified with good performance in callus induction and plant regeneration.
