摘要
目的:研究狼紫草水提物对小鼠皮肤溃疡组织环氧酶-2(cyclooxygenase-2,COX-2)表达的影响。方法:煎煮法制备狼紫草水提物,醋酸化学刺激法制备小鼠体表溃疡模型。分别采用Western blotting和RT-PCR法测定局部皮肤溃疡组织中环氧酶-2蛋白及其mRNA表达情况。结果:正常小鼠皮肤组织中几乎无COX-2表达,模型组小鼠体表溃疡组织中COX-2蛋白及其mRNA的表达均显著增加,应用高剂量(40g.kg-1)和中剂量(20g.kg-1)狼紫草水提物治疗后,二者表达水平明显降低。结论:狼紫草水提物对COX-2有明显的干预作用,可预见其抗炎镇痛作用有较好应用前景。
OBJECTIVE To explore the effects of aqueous extract from Lycopsis orientalis L.（AELO） on the expression of COX-2 in skin ulcer tissue of mice.METHODS AELO was prepared with decocting method and the skin ulcer model was induced by acetic acid chemical stimulus.The expression of cyclooxygenase（COX）-2 protein and mRNA were detected with Western blotting and RT-PCR technique respectively.RESULTS The expressions of COX-2 protein and mRNA obviously increased in skin ulcer tissue as compared with those of normal control group.Nevertheless,after the administration of AELO（40 or 20 g·kg-1）,their expressions decreased significantly.CONCLUSION AELO can significantly down-regulate the expression of COX-2 protein and mRNA in skin ulcer tissue.
