摘要
目的探讨高糖环境对大鼠肾小球系膜细胞增殖、单核细胞趋化蛋白-1(MCP-1)的表达及NF-κB活化的影响。方法将体外培养大鼠肾小球系膜细胞(MC)分为4组:正常对照组(含5.6 mmol/L葡萄糖)、高糖1组(含15 mmol/L葡萄糖)、高糖2组(含20 mmol/L葡萄糖)和高糖3组(含25 mmol/L葡萄糖)。MTT法检测细胞的增殖情况,ELISA法检测MCP-1的表达,Western blotting法检测IκBα以反映NF-κB的活性。结果与正常对照组比较,高糖环境下24 h后细胞增殖明显,MCP-1表达明显增强(P<0.01)。高糖刺激30 min后,NF-κB结合蛋白IκBα明显降解,提示高糖可诱导MC NF-κB活化。结论高糖环境下大鼠肾小球系膜细胞的增殖及炎症因子MCP-1的高表达可能是通过NF-κB的活化来实现的。
Objective To explore the effect of high glucose on cells proliferation,monocyte chemoattractant protein-1 (MCP- 1) expression and NF-KB activation in rat mesangial cell(MC). Methods The in vitro culture of rat glomerular mesangial ceils were divided into four groups: normal glucose group (glucose concentration of 5.6 retool/L), high glucose group 1 (glucose concentration of 15 mmol/L), high glucose group 2 (glucose concentration of 20 mmol/L), and high glucose group 3 (glucose concentration of 25 retool/L). MTI' method was used to detect cell proliferation. MCP-1 in the supernatant was measured by ELISA. IKBct expression was detected by Western blotting method to reflect the NF-KB activation. Results The MC proliferation and MCP-I expression were significantly increased after 24 h in high glucose groups (P〈0.01). The expression of NF-KB-binding protein IKBct was obviously declined after 30 min of high glucose treatment, and indicated that high glucose could induce the NF-KB activation in MC. Conclusion High glucose could stimulate rat glomerular mesangial cell proliferation and promoted inflammation factor MCP-1 expression may be through the NF-KB activation.
