摘要
以优化少根根霉液体深层发酵技术为目标,利用经过预先优化的培养基配比,研究搅拌速率、通气量、初始pH对发酵效果的影响。结果表明优化的发酵条件为:搅拌速率200 r/min,通气量5.63 L/min,初始pH 6。在优化的条件下进行10 L发酵罐验证试验,发酵时间为96 h时,少根根霉菌丝体生物量达到3.75 g/L,生物转化率达到8.21%,油脂得率为26.5%。
Using optimized medium, the influences of stirring speed, ventilation volume and initial pH on fermentation effect were studied so as to optimize fermentation conditions in submerged culture of Rhizopus arrhizus. The results showed that the optimal fermentation conditions were as follows:stirring speed 200 r/min, ventilation volume 5.63 L/min, initial pH 6. Under the optimal conditions, the highest biomass of Rhizopus arrhizus mycelium was 3.75 g/L, biological transformation rate was 8.21% and lipid yield was 26.5% when Rhizopus arrhizus fermented in 10 L fermentation tank for 96 h.
