摘要
目的 用HPLC检测人血清中的特布他林浓度。方法 样品用硅胶萃取小柱纯化浓集后用C8柱 (5 μm ,2 5 0mm× 4.6mm)作分析柱 ,用硫酸沙丁胺醇作内标 ,以pH2 .8,0 .0 3mol·L- 1 NH4H2 PO4缓冲液 CH3OH CH3CN(90∶3∶7)为流动相 ,在波长为2 2 4nm处紫外检测。结果 标准曲线在 0 .5～ 32 μg·L- 1 范围内有良好线性关系 (r =0 .9998) ,最低检测浓度为 0 .2 μg·L- 1 ,平均方法回收率为 (10 0 .7± 3 .1) % ,日内RSD小于 3.7% ,日间RSD小于 7.5 %。结论 方法简便 ,快速准确 。
OBJECTIVE To determine the concentration of Terbutaline in human serum by HPLC with solid phase extraction.METHODS The extract of serum sample was eluted by YWG C 8 (5 μm,250 mm×4.6 mm) column with 0.03 mol·L -1 phosphate buffer (pH2.8) methanol acetonitrile (90∶3∶7) as the mobile phase,and detected at 224 nm,Albuterol was used as the internal standard.RESULTS The retention times of Terbutaline and Albuterol were 7.9 min and 7.4 min,respectively.The standard curve of Terbutaline was linear within the concentration range of 0.5 to 32.0 μg·L -1 ( r =0.9998).The detection limit was 0.2 μg·L -1 .The within day RSD (%) was less than 3.7,and the inter day RSD (%) less than 7.5.The average recovery of Terbutaline was (100.7±3.1)%.CONCLUSION This method was found to be simple,quick and accurate for the determination of Terbutaline in human serum.
