不同时间段的预变性神经段分子生物学变化 被引量:1

Molecular biological changes withina pre-degenerated nerve

摘要 目的 了解预变性神经移植修复神经缺损较新鲜神经移植修复效果良好的机制 ,并研究预变性神经段分子生物学变化。方法 选用 3 6只SD大鼠 ,将大鼠双侧上肢尺神经在腋部切断 ,远断端埋入肱二头肌中。术后当天、3d、1周、2周、3周、4周 ( 1~ 6组 )取材 ,应用rt PCR ,SouthernBlot的方法检测不同时间段的预变性神经中BDNF和CNTF的变化。结果 预变性神经中BDNF和CNTF的变化有明确的特点。预变 1周 (第 3组 )内BDNF的含量最高 ,以后 ,在较长的时间内 ( 14~ 2 8d ,即第 4,5 ,6组 )维持在较高水平。CNTF的含量有两个高峰 ,一个高峰是在预变后第 3天 (第 2组〕 ,第二个高峰是在预变后 3周 (第 5组 ) ,此后CNTF又下降。结论 Schwanncells在不同时间段的预变性神经段分泌不等量BDNF和CNTF ,两者的协同作用可促进神经再生。 Objective To study the temporal molecular biological changes within the pre degenerated nerve, and explore the mechanism of the “pre degenerated” phenomenon.Methods The ulnar nerves were cut bilaterally at the axillary level in 36 SD rats. The distal end of the nerve was buried in the biceps brachii muscle. Samples of the pre degenerated nerves were harvested right after the operation, 3 days, 1 week, 2 weeks, 3 weeks, and 4 weeks postoperatively (group 1 6) to examine BDNF and CNTF level at each interval using rt PCR and Southern Blot.Results BDNF and CNTF change in the pre degenerated nerve was characteristic. BDNF level was highest 1 week after degeneration (group 3). The high level of BDNF maintained for a certain period (14 to 28 days). The level of CNTF had two peaks. One peak was in group 2 in which the nerve was pre degenerated for 3days. The second peak was in group 5 with 3 weeks' pre degeneration. CNTF level decreased afterwards.Conclusions Schwann cells in the nerves of different pre degenerative intervals secret different levels of BDNF and CNTF. The synergetic effect of the two factors promotes nerve regeneration.
