金丝桃苷对高糖和氧化损伤所致内皮细胞凋亡的影响 被引量:5

Effects of Hyperoside on the Apoptosis of Endothelial Cells Due to High Glucose and Oxidative Damage

摘要 目的:研究金丝桃苷(HP)对高糖和氧化损伤所致内皮细胞凋亡的影响。方法:体外培养内皮细胞,分为正常细胞、正常细胞+HP、高糖损伤、高糖+HP、H2O2损伤与H2O2+HP组,采用MTT法测定内皮细胞的活力,并采用RT-qPCR法、Westernblot法分别测定B细胞淋巴瘤/白血病-2基因(Bcl-2)蛋白及其mRNA表达水平,Bcl-2相关X蛋白(Bax)及其mRNA表达水平。结果:与各自损伤组比较,高糖+HP、H2O2+HP组内皮细胞损伤程度显著减轻(P<0.01),高糖+HP、H2O2+HP组内皮细胞Bcl-2mRNA和蛋白表达显著增强(P<0.01),BaxmRNA和蛋白表达显著减弱(P<0.01)。结论:HP通过抑制Bax的表达,上调Bcl-2的表达而拮抗高糖、氧化损伤导致的内皮细胞凋亡,对糖尿病性视网膜病变血管内皮细胞具有一定的保护作用。 OBJECTIVE: To study the effects of hyperoside(HP) on the apoptosis of endothelial cells due to high glucose and oxidative damage. METHODS: Endothelial cells were cultured in vitro and divided into normal cells, normal+HP, high glucose in- jury, high glucose+HP, H202 injury and H202 injury +HP groups. MTT assay was used to determine the activity of endothelial cells, and RT-qPCR method and Western blot assay were adopted to detect mRNA and protein expression of Bcl-2 and Bcl-2 related X protein (Bax). RESULTS: Compared with injury group, the injury of endothelial cells relieved significantly in high glucose+ HP group and H20~ injury +HP group(P〈0.01 ). mRNA and protein expression of Bcl-2 increased significantly in high glucose+HP group and H202 injury +HP group (P〈0.01), while the mRNA and protein expression of Bax decreased significantly (P〈0.01). CONCLUSION: By inhibiting the expression of Bax and up-regulating the expression of Bcl-2 , HP can antagonize apoptosis of en- dothelial cells due to high glucose or oxidative damage. HP has a orotective effect on DR vascular endothelial cells.
