摘要
采用 Bom氏比浊法测定灯盏花总黄酮 ( Erigeron breviscapus flavones,EBF)在体内和体外对血小板活化聚集的影响。应用 Kohler等法观察 EBF对小鼠尾静脉注射花生四烯酸 ( AA)所致突然死亡的保护作用。结果表明 ,EBF在体外和体内对 ADP、AA或血小板活化因子 ( PAF)引起的血小板聚集均有明显抑制作用 ,体外实验 ,其效应呈浓度依赖关系 ,体内试验 ,于给药后 180 min达最大抑制效应 ,药效持续 2 4 0 min以上 ;EBF显著对抗 AA引起的体内血栓形成作用 ,呈剂量—效应相关性 ,结果揭示 ,EBF有利于防治伴随血小板聚集性增高的血栓栓塞性疾病。
Effects of erigeron breviscapus flavones(EBF)on platelet aggregation.were determined by Born’s method;the method of Kohler et al .was used to evaluate the prevention of EBF against the mouse sudden death caused by i.v.injection of arachidonic acid(AA).The results showed that EBF in vitro significantly inhibited ADP,AA and platelet activating factor(PAF) induced platelet aggregation in a concentration dependent manner;50 mg/kg of EBF produced markedly inhibitory effects( P < 0.05 )15 min after oral administration,showed maximal effects at 180 min and maintained its action for over 240 min.In thrombosis caused by i.v.injection of AA,EBF dose dependently suppressed the mortality of mice.It is suggested that EBF is advantageous to prevention and therapy of thromboembolic diseases especially in combination with hyperfunction of platelet aggregation.
