rIL-10诱导下子宫腺肌病患者子宫内膜细胞HLA-G的表达变化 被引量:2

摘要 目的:探讨子宫腺肌病患者的子宫内膜细胞在重组IL-10(rIL-10)刺激下,HLA-G的表达变化。方法:取25例因子宫腺肌病切除子宫患者的子宫内膜分离腺上皮细胞及间质细胞行体外培养,采用角蛋白及波形蛋白分别鉴定两种细胞纯度,通过单纯rIL-10干预培养和rIL-10、IL-10受体(IL-10R)抗体联合干预培养,采用免疫细胞化学SP法及Western bolt技术检测子宫内膜腺上皮细胞在干预前后HLA-G分泌的变化情况;采用流式细胞学技术及免疫细胞化学SP法检测子宫内膜间质细胞在干预前后HLA-G分泌的变化情况。结果:rIL-10干预48 h后,单纯rIL-10干预组(干预组B)子宫内膜腺上皮细胞HLA-G的表达量增加(P<0.05),IL-10R抗体及rIL-10联合干预组(干预组A)以及无rIL-10干预组(未干预组)HLA-G的表达量无明显变化(P>0.05)。干预组B及干预组A子宫内膜间质细胞HLA-G的表达量减少(P<0.05),未干预组子宫内膜间质细胞HLA-G的表达量无明显变化(P>0.05),间质细胞阳性表达率无明显变化(P>0.05)。结论:rIL-10刺激子宫腺肌病患者的子宫内膜腺上皮细胞HLA-G的表达量增加,rIL-10刺激子宫腺肌病患者表达HLA-G可能是通过IL-10Rα起作用的,rIL-10诱导HLA-G的产生使子宫腺肌病患者异位子宫内膜产生免疫抑制作用。 Objective: To explore the change of expression of human leukocyte antigen-G (HLA-G) via recombinat interleukin-10 (rIL-10) induction in stromal cells and glandular epithelium cells of patients with adeno- myosis. Methods: Laboratory study using the human endometrium stromal cells and glandular epithelium cells ob- tained from 25 patients with adenomyosis undergoing hysterectomy. HLA-G protein expression in two cells via IL-10 induction with Flow cytometry assay, Western blotting and Immunofluorescence. Quantitative analysis of HLA-G protein expression via IL-10 induction, according to Student's t-test. Results: After the intervention of the rIL-lO for 48 h, a simple intervention group of rIL-lO ( intervention group B) , the HLA-G expression of endometrial glan- dular epithelial ceils was increased ( P 〈 0. 05 ) IL-IOR antibody and rIL-lO joint intervention Group ( intervention group A) and rIL-10 intervention group (not intervention group) have no significant change in the amount expres- sion of HLA-G(P 〉 0. 05 ). Compared with stromal cells without IL-10 induction, HLA-G protein expression had a statistically significant decrease in these with IL-10 induction ( P 〈 0. 05 ), but it is opposite for glandular epithelium cells. After IL-10 induction, the positive expression rate of HLA-G expression in two cells did not have obviously change (P 〉 0. 05 ). Conclusion: rIL-10 stimulation increased the HLA-G expression of adenomyosis endometrial glandular epithelial ceils in patients, rIL-10 stimulation in patients with adenomyosis expression of HLA-G may be the role of be the role of IL-10 Rα which can make adenomyosis patients with ectopic endometrium induce immune inhibition
