摘要
分别从3组不同日龄鸽消化道(口腔、嗉囊及内脏)采集毛滴虫特征性病变内容物,经处理后接种在健康鸽肝脏组织块原代外植pH7.0培养液中,置37.5℃培养。250倍光学显微镜观察,虫体在24~36h生长达到高峰,数量为200~517个/μL,大量虫体定殖肝组织块中,并进行阿米巴样快速游动,同时可见数百虫体黏附聚集瓶底绞结成团及大量体积增加两倍的分裂体。5d后在瓶底可见死亡虫体,7d后活虫体数量逐渐下降,12d活虫体减至20个/μL,培养基液pH6.0。每隔7d传代培养,最多可传至F10代。
The contents from digestive tracts (mouth, gizzard, internal organs) that showing lesions of suspective Trichomonas gallinae infection were taken from 3 groups of pigeons at different ages. Samples after treatment were inoculated into healthy pigeon liver block primary explantation medium at pH 7.0 and incubated at 37.5℃. The culture smear observed under 250× optical microscope,the polypide reached maximum growth at 24 to 36h with 200 to 517 polypides/μL. A large amount of polypides localized in the liver and showed amoeboid rapid movement. Meanwhile hundreds of polypides were adhered to the bottom of the bottle and formed lumps with the size of two times more. Five days later dead polypides could be seen at the bottom of the bottle and 7 days later more polypides were settled down gradually and by 12 days live polypides reduced to 20 per μL, and medium pH dropped to 6.0. The polypides had been passaged every 7 days,and they could only be passaged for 10 generations.
