灯台叶醇提物对C57BL/6荷瘤小鼠免疫功能的影响 被引量:4

摘要 目的研究灯台叶醇提物对C57BL/6荷瘤小鼠免疫功能的影响。方法建立C57BL/6雄性小鼠腋下皮下注入Lewis肺癌小鼠移植瘤动物模型,将荷瘤鼠分为:正常组,荷瘤小鼠组,灯台叶醇提物低、中、高剂量组(0.2,1.0,5.0g.kg-1),灌胃给药,连续给药14d,分别测定其抑瘤率、T淋巴细胞亚群CD4+/CD8+;小鼠血清中白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、干扰素-γ(INF-γ)水平。结果灯台叶醇提物高剂量组对肿瘤生长有明显的抑制作用(P<0.05),其抑瘤率为64.9%;中、高剂量组能显著提高荷瘤小鼠血清INF-γ、IL-6以及IL-10的水平(P<0.05);高剂量组对小鼠胸腺质量增加无明显影响,但抑制脾脏增大;能够降低CD4+/CD8+比例,并且增强T淋巴细胞增殖能力(P<0.05)。结论灯台叶醇提物具有提高荷瘤小鼠免疫功能的作用。 Objective To study the immunoregulating effect of the ethanol extract of Alstonia scholaris(AS) on C57BL/6 tumor-bearing mice.Methods Lewis cells were injected subcutaneously into C57BL/6 male mice to make xenograft animal models.Tumor-bearing mice were divided into normal group,tumor-bearing mice group,AS low,medium and high dose groups(0.2,1.0 5.0 g·kg-1) by intragastric administration for 14 consecutive days.T lymphocyte subsets of CD4+/CD8+;interleukin-6(IL-6),leukocyte interleukin-10(IL-10),and interferon-γ(INF-γ) level in the tumor-bearing mice serum were measured to investigate the tumor inhibitory rate.Results The high-dose of AS exhibited significant inhibitory effect on tumor growth(P0.05),and the inhibition rate was 64.9%.Medium and high dose groups significantly improved the INF-γ,IL-6,and IL-10 levels in tumor-bearing mice serum(P0.05).High-dose group had no significant effect on the mouse thymus weight,but inhibited splenomegaly.High-dose group was able to reduce the CD4+/CD8+ ratio and to enhance T lymphocyte proliferative capacity(P0.05).Conclusion AS has a role to improve the immune function of tumor-bearing mice.
