增骨Ⅲ号影响Ⅰ、Ⅱ型胶原基因表达的实验研究 被引量:12

摘要 目的 从分子水平探讨增骨Ⅲ号的作用机理。方法 取 12 0只昆明小鼠造成桡骨1 5mm缺损骨折 ,随机分为 2组 :服药组及未服药组 ,各 6 0只 ,在术后 2周和 4周 ,分别进行骨痂总RNA的分离提取 ,通过与Ⅰ、Ⅱ型胶原质粒探针 (pMCol1a 1及 pMCol2a 1)斑点杂交 ,比较Ⅰ、Ⅱ型胶原mRNA表达的变化。结果 Ⅰ型胶原mRNA表达在术后 2周明显小于 4周 ,Ⅱ型胶原mRNA表达在术后 2周明显大于 4周 ,提示Ⅰ型胶原mRNA表达代表骨形成及塑形的特征标志 ,Ⅱ型胶原mRNA表达代表软骨修复的特征标志。Ⅰ型胶原mRNA表达量 ,服药组大于未服药组 ;Ⅱ型胶原mRNA表达量 ,服药组小于未服药组。提示服药组骨形成迅速 ,由软骨修复期提早进入骨修复及塑形期。结论 增骨Ⅲ号能促进及加快骨的修复。 Objective [HT5”SS]CH(Chinese Herbal)Zeng-gu No.Ⅲ is one of a serial medicines for the treatment of osteoporosis;gene technology is applied in this experiment for the investigation of its molecular mechanism.[HT5”H]Methods [HT5”SS]Models of radial fracture with bone defects of 1 5 mm were produced in 120 mice.The animals were divided into 2 groups randomly of the treatment and control groups.At the 2nd week after operation,total RNA in callus of fracture site of the two groups were extracted and were hybridized with probes of pMCol1a-1 and pMCol2a-1 in dot blot.The changes of mRNA expression of type Ⅰ and type Ⅱ collagen were compared.[HT5”H]Results [HT5”SS]Whether taking CH or not,the expression level of mRNA of type Ⅰ collagen at the 2nd week after operation was apparently lower than that in the 4th week,however,that of type Ⅱ collagen at the 2nd week after operation was apparently higher than that at the 4th week.The experiment suggested that the expression of mRNA of type Ⅰ collagen was the marker of bone formation and bone mouldling,while that of type Ⅱ collagen is the marker of cartilaginous repair.Compared with treatment control group,the mRNA expression level of type Ⅰ collagen is lower,but that of type Ⅱ collagen was higher.Apparently osteogenesis in treatment group increased rapidly.It took less time to finish the change from cartilaginnous repair stage to bone repair and moulding stage.[HT5”H]Conclusion [HT5”SS]Zeng-gu No.Ⅲ could enhance and increase bone repair. [WT5”HZ]
