Ⅰ型糖尿病患者外周血中骨桥蛋白与IL-12的表达及临床意义 被引量:2

The expression and clinical significance of osteopontin and IL-12 in diabetes

摘要 目的探讨Ⅰ型糖尿病患者外周血中骨桥蛋白(OPN)与IL-12的表达及临床意义。方法选取2011~2012期间于本院诊治的Ⅰ型糖尿病患者50例。并收集同期健康体检者50名作为对照组。采用ELISA法检测血浆中OPN和IL-12的水平,并运用实时荧光定量RT-PCR检测外周血单个核细胞中OPN和IL-12基因的表达水平。结果Ⅰ型糖尿病患者外周血中OPN与IL-12水平均高于对照组(t值分别为40.7、33.7,P<0.05)。Ⅰ型糖尿病患者外周血中单个核细胞中OPN与IL-12mRNA表达水平与对照组比较,差异有统计学意义(P<0.05)。Ⅰ型糖尿病患者外周血中OPN与IL-12水平呈正相关(r2=0.936,P=0.00)。结论Ⅰ型糖尿病发生与骨桥蛋白和IL-12表达有关,骨桥蛋白与IL-12水平呈正相关。 Objective To study the expression of osteopontin(OPN)and interleukin-12and analyze their roles in the pathogenesis of diabetes.Methods 50patients with diabetes typeⅠdiagnosed and treated in the hospital from 2011to 2012were enrolled,and 50health controls were also enrolled from medical examination center during the same period.OPN and IL-12levels were measured by ELISA kits.The mononucluear cells were collected from peripheral blood,and the expression levels of OPN and IL-12gene were detected by real time fluorescent quantitative reverse transcription polymerase chain reaction.Results In diabetes group,levels of OPN and IL-12were markedly higher than those of healthy control(t=40.7,33.7respectively,P0.05).The gene expression level of OPN and IL-12mRNA were higher than conrol group(P0.05).IL-12were positively correlated with OPN levels(r2= 0.936,P=0.00).Conclusion OPN and IL-12levels in peripheral blood of diabetes patients are closely related to the diabetes degree.
