AEG-1-shRNA阻断AEG-1对肝癌细胞BEL-7402侵袭性的影响 被引量：3

摘要 目的探索星形细胞上调基因(AEG)-1-shRNA阻断AEG-1对肝癌细胞BEL-7402侵袭性的影响。方法应用pU6mRFP AEG-1-shRNA阻断肝癌细胞BEl-7402的AEG-1表达,设立阴性质粒对照组和空白对照组。半定量RT-PCR法比较侵袭相关基因金属基质蛋白酶(MMP)2、MMP9、血管内皮生长因子(VEGF)mRNA的表达;Millicell检测细胞侵袭能力。结果 pU6mRFP AEG-1-shRNA组与对照组相比AEG-1的表达显著降低(P﹤0.05);细胞侵袭能力下降,侵袭相关基因MMP2、MMP9、VEGF表达减少(P﹤0.05)。结论 shRNA沉默AEG-1明显抑制了人肝癌细胞BEL-7402的侵袭性及其相关基因的表达,AEG-1是有效治疗肝细胞癌的靶点。
