摘要
采用组织块贴壁法,对鲤鱼的吻端和尾鳍进行细胞的离体培养,观察所获得细胞的形态,并总结每种细胞的形态学特点,同时绘制F5代细胞的生长曲线.研究结果表明,在原代细胞培养过程中,鲤鱼吻端细胞生长较快,以上皮样细胞为主,而鲤鱼尾鳍细胞生长较慢,以成纤维细胞居多.传代培养发现,通过控制胰蛋白酶消化的时间,尾鳍组织经2~3代筛选后可获得较纯的成纤维细胞.从F5代生长曲线看出,鲤鱼尾鳍细胞增殖能力略强于吻端细胞,因此鲤鱼尾鳍组织是较适宜的成纤维细胞体外培养材料.
In the process of cells culture,the morphology and proliferation quality of cultured cells is important and it affects other related technologies.In this paper,snout and tail fin tissues of carp were cultured in vitro by tissue adhesion method and the morphology of the obtained cells was studied.We summarized the morphological characteristics of each cell and drew the growth curve of F5 cells.The results showed that in primary cell culture,the carp snout cells grow faster,mainly were skin-like cells.Carp fin cells grow more slowly,mainly were fibroblasts.In the process of cells subculture,we found that more pure fibroblasts were obtained after the caudal fin tissues were screened for 2 to 3 generations by controlling the time of trypsin digestion.The F5 growth curve indicated that proliferation of carp fin cell was slightly stronger than that of the snout cell.This result suggested that carp tail organization was more suitable material for fibroblasts culture in vitro.
