神经组织联合移植修复成鼠脊髓损伤的比较研究 被引量:7

摘要 目的 比较胚胎脊髓与带蒂或游离周围神经联合移植修复脊髓损伤的能力。 方法  30只成年大鼠损伤胸髓后行胚胎脊髓加游离正中神经 (P +F组 )或加带蒂正中神经 (V +F组 )移植。术后 8周行体感诱发电位 (SEP)和光、电镜检查。 结果 V +F组胚胎脊髓与受体融合较好 ,体积增长 378.4%、神经微丝蛋白密度 (7.46± 0 .77)万个 /mm2 、神经元密度 (1.36± 0 .34)万个 /mm2 ,均高于P +F组 (P <0 .0 5 ) ,细胞分化程度高 ,SEP的P1、N1波潜伏期显著缩短 (P <0 .0 5 )。 结论 胚胎脊髓与带蒂神经联合移植 ,能促进胚胎脊髓成熟分化和损伤神经元再生 。 Objective To compare the effects of combined fetal spinal cord allograft and vascularized or free peripheral nerve autograft to repair spinal cord injury in adult rats. Methods A total of 30 adult, female Wistar rats were used in this study, and a 5 mm cord defect of the left lateral column was made at T 1~3 vertebral level. The defect was grafted randomly using embryonic day 14 fetal spinal cord and vascularized (Group V+F), or free peripheral nerve(Group P+F) respectively. Eight weeks after surgery, the survival, differentiation of the grafts, and the ability of repairing host spinal cord were assessed by histological, histomorphometric analysis and somatosensory evoked potential (SEP) studies. Results In group V+F, the fetal implant was intimately fused with the host CNS parenchyma. The volume, nerve fiber and neurons density of the fetal implant were significantly higher than Group P+F ( P <0.05). Grafted fetal spinal cord exhibited a high level of maturity, SEP assessments showed that the latency of P 1, N 1 wave was significantly decreased in this group ( P <0.01). Conclusions Group V+F has an advantage over group P+F in histological and SEP, and this combination could promote the development of fetal implant and support the regeneration of central injured axons to certain degrees.
