响应面法优化玉竹多糖的酶法提取工艺 被引量:5

Optimization of Enzymatic Extraction of Crude Polysaccharide from Rhizoma Polygonati Odorati by Response Surface Methodology

摘要 【目的】优化酶法提取玉竹多糖的工艺。【方法】在单因素实验的基础上,采用响应面分析法对影响酶法提取玉竹多糖得率的主要因素(酶解时间、酶解温度和酶用量)进行优化,建立了影响因素与玉竹多糖得率之间的函数关系。【结果】玉竹多糖酶法提取的最佳工艺为:木瓜蛋白酶加酶量为6.5 g/L,酶解温度为50℃、酶解时间为125 min,在此条件下,玉竹多糖得率达到100.89 mg/g,试验结果与模型预测值相符。【结论】Box-Behnken设计结合响应面分析法可以很好地对玉竹多糖酶法提取工艺进行优化。 Objective To optimize enzymatic extraction process of crude polysaeeharide from Rhizoma Polygonati Odorati. Methods Response surface methodology (RSM) was used to optimize the enzymatic extraction of crude polysaccharide from Rhizoma Polygonati Odorati based on single-factor experiment. The influences of main factors of extraction temperature, extraction time and enzyme concentration (mass fraction) on the process were observed. The relationship between influencing factors and response values was investigated. Results The optimal conditions for the enzymatic extraction were extraction for 125 min at 50 ℃ with the 6.5 g/L of enzyme. Under the optimized conditions, the maximum yield of polysaccharides reached 100. 89 mg/g , which matched well with the predicted value. Conclusion Box-Behnken design combining with response surface analysis is effective for the optimization of the enzymatic extraction of crude polysaccharide from Rhizoma Polygonati Odorati.
