摘要
目的 观察巨噬细胞 (MΦ)及转化生长因子 β1(TGF β1)在胆道愈合过程中的表达和分布 ,探讨其在良性胆管狭窄形成过程中的作用及意义。方法 通过制作犬胆管损伤修复模型 ,术后分为 1周组、3周组、3个月组、6个月组。采用免疫组织化学过氧化酶标记的链霉卵白素 (SP)法对MΦ、TGF β1在各组中的表达和分布进行研究。结果 MΦ 1周时表达 ,与其他各期比较差异有显著性(P <0 .0 5 ) ,3周~ 6个月表达差异无显著性 (P >0 .0 5 ) ;TGF β1愈合过程中持续表达较强 ,各期比较差异无显著性 (P >0 .0 5 )。MΦ主要表达于粘膜固有层 ;TGF β1主要表达于肉芽组织、成纤维细胞、MΦ及血管内皮细胞。结论 MΦ、TGF β1高表达是造成胆道愈合过程成纤维细胞增殖旺盛、细胞外基质过度沉积、瘢痕增生的重要因素。
Objective\ To observe the expression of transforming growth factor β 1 (TGF β 1 ) and distribution of macrophage in the healing process of bile duct and discuss their roles and implication in the process of benign biliary stricture formation. Methods\ A repairing model of trauma of bile duct in the dog was made. The dogs were divided into one week group, 3 week group, 3 month group and 6 month group. The expression of TGF β 1 and distribution of macrophages in the tissue in the different groups after operation were studied by using immunohistochemical SP staining method. Results\ High expression of macrophages and TGF β 1 was observed in the healing process and lasted for a long time. Macrophage staining was strong positive in the one week group, significantly different from that in the other groups ( P < 0.05 ). Macrophage staining was found at the intrinsic layer under mucosa membrane, TGF β 1 staining at the granulation tissue, fibroblasts and endothelial cells of blood vessels. Conclusion\ The high expression of macrophages and TGF β 1 was related closely with the fibroblast proliferating actively, extracellular matrix overdeposition and scar proliferation in the healing process of bile duct.\;
