香菇多糖对约氏疟原虫感染小鼠树突状细胞的免疫调节作用 被引量:5

Immunoregulation of dendritic cells by lentinan in mice infected with Plasmodium yoelii

摘要 目的观察香菇多糖(lentinan,Lent)对约氏疟原虫(Plasmodium yoelii 17XL,Py17XL)感染小鼠树突状细胞(dendritic cells,DCs)的免疫调节作用。方法 120只BALB/c小鼠随机分为实验组和对照组,腹腔接种1×106Py17XL寄生的红细胞后,实验组用香菇多糖腹腔注射处理,对照组仅采用PBS腹腔注射处理,分别统计小鼠的平均生存时间和原虫血症水平;分离小鼠脾脏DCs细胞,用Western blot检测其IL-2、IL-10、IL-12、TGF-β、CD86和TLR-9蛋白表达水平。结果实验组和对照组小鼠平均生存时间为(8.35±2.55)d和(5.30±1.26)d,差异有统计学意义(P<0.05)。实验组脾脏DCs细胞IL-2、IL-12、TGF-β、CD86和TLR-9蛋白表达水平显著高于对照组(P<0.05),IL-10蛋白表达水平显著低于对照组(P<0.05)。结论香菇多糖具有抗疟作用,能通过上调IL-2、IL-12、TGF-β、CD86和TLR-9等细胞因子水平促使DCs成熟和加强抗原递呈作用。 Objective To observe the immunoregulation of dendritic cells by lentinan in mice infected with Plasmodium yoelii. Methods A total of 120 female BALB/c mice were inoculated with red blood cells infected with 1 × 106 PylTXL P. yoelii and were randomly divided into an experimental group and control group. The experimental group was intraper itoneally injected with lentinan, and the control group was injected with PBS. The average survival time of mice and para site levels of viremia were determined, and the levels of IL-2, IL-10, IL-12, TGF-β, CD86, and TLR-9 were detected in mouse spleen DCs using Western blotting. Results The average survival time was (8.35±2.55) days for the experi mental group and (5.30±1.26) days for the control group; the difference was significant (P〈0.05). The experimental group had higher levels of expression of IL-2, IL-12, TGF-β, CD86, and TLR-9 in spleen DCs than did the control group; the difference was significant (P〈0.05). The experimental group had higher levels of IL-10 expression than did the control group; the difference was significant (P〈0.05). Conclusions Lentinan acts against malaria, furthering the maturation and enhancing the antigen-presenting role of dendritic cells by up-regulating the levels of IL-2, IL- 12, TGF-β, CD86 and TLR-9.
