摘要
为开展绿木霉ZBS6(Trichoderma viride ZBS6)拮抗基因筛选及克隆工作,建立了高质量基因组DNA的提取方法,并采用SuperCos1载体构建了绿木霉ZBS6黏粒基因组文库。试验中共获得3.9×104个黏粒克隆,平均插入片段大小约为33kb,文库滴度约为4.2×108 cfu/mL。理论上,从该文库中筛选到任一DNA序列的精确概率可高达99.9%。
To cope with the screening and cloning of antagonistic genes of Trichoderma viride ZBS6,this study established a method for obtaining high-quality genomic DNA and constructed the genomic library of T. viride ZBS6 by SuperCosl vector. The genomic library consisted of 3.9× 10^4 independent clones with an average insert size of about 33 kb, and had a colony titer of around 4.2 ×10^8 cfu/mL in stock. So the exact probability of having any given DNA sequence in the library was as high as 99.9%.
