应用基因芯片筛查鼻息肉相关差异表达基因 被引量:1

摘要 目的应用基因表达谱芯片筛查鼻息肉相关差异表达基因。方法采用含有人类全长基因的寡聚核苷酸芯片检测6例鼻息肉组织、6例正常鼻黏膜组织的相关差异表达基因,选择部分差异表达基因进行RT-PCR验证。结果筛查出鼻息肉组织差异表达基因共1886条,共同差异表达基因18条,上调基因12条,下调基因6条,包括编码横跨膜4超家族蛋白、IFN-γ诱导蛋白IP-30前体、补体因子前体、胰岛素生长因子结合蛋白、IL-8、RGS1、GRRK4、CCL20、子宫球蛋白等基因。RT-PCR测定差异表达基因IL-8、RGS1与基因芯片检测结果一致。结论鼻息肉基因表达谱存在差异,RGS1通过影响细胞信号传导通路发挥作用,IL-8促使炎性细胞释放多种炎性递质参与鼻息肉发病,IFN-γ诱导蛋白和胰岛素生长因子结合蛋白等基因在鼻息肉发病中起重要作用。 Objective To explore the changes in gene expressions in human nasal polyp(NP). Methods In this study, microarrays containing individual full length DNA from homo sapiens were applied to screen differentially expressed genes in 6 NP samples and 6 normal mueosal samples. The differentially expressed genes were identified and subjected to real time PCR analysis. Results One thousand eight hundred and eighty six( 1 886) genes were differently expressed in gene profile of NP. Eighteen differentially expressed genes were found in NP samples and normal mucosal samples, among which 12 genes were up-regulated and 6 were down-regulated. The differentially expressed genes mostly involved in cytokines, adhesion genes about complements and their receptors, immune transcription regulatory molecules and signal transduetion included IL-8, RGS1, GRK4, CCL20, uteroglobin, protein of the transmembrane 4 superfamily, highly similar to GAMMA interferon inducible protein IP-30 precursor, highly similar to complement factor I precursor and insulin like growth factor binding protein 3. The real time PCR results of IL-8, RGS1 were consistent with those of gene chip analysis. Conclusions Mieroarray analysis was efficient in the screening of differentially expressed genes related to the human NP. RGS1 might play an important role in cell signal transduction. IL-8 might involve in the occurrence of NP by inducing the release of inflammatory factors. NP was a polygenic disease and the genes of GAMMA interferon inducible protein, insulin like growth factor binding protein might play an important role in its pathogenesis.
