摘要
目的 :为了解本地区原发性肝癌 (PHC)中HGV感染状况和与输血的关系。方法 :采用逆转录 套式聚合酸链反应法 (RT nPCR)对 2 0例PHC和 4 4例职业献血员进行了HGV RNA检测 ,并对 3例阳性产物纯化克隆后 ,采用ABIPrism 377DNA自动测序仪 ,对其 5′非编码区基因序列 ( 115~ 4 67nt)进行了测定。结果 :2 0例PHC中 ,2例 ( 10 .0 % )HGV RNA阳性 ,该 2例分别为HBV、HCV、HGV三重感染和HBV、HGV重叠感染 ,且均有输血史 ;4 4例职业献血员中 ,1例 ( 2 3% )HGV RNA阳性。该 3株HGV核苷酸序列同源性为 97.3%~ 98.8% ,与GBV C(U36380 )、HGV (U4 4 40 2 )和HGV3型 (D90 60 1)的核苷酸同源性分别为 88.3%~ 89.4 %、88.4 %~ 89.7%和 95.2 %~ 96.3%。结论 :PHC中HGV感染与输血有关 ;HGV可能与PHC的发生无关。本地区HGV感染株属基因
Objective:To investigate the status of hepatitis G virus(HGV)infection.Method:HGV RNA was tested by RTnPCR among 20 patients with primary hepatocellular carcinoma (PHC)and 44 paid blood donors.Results:The positive rates of HGV RNA in PHC and blood donors were 10.0%(2/20) and 2.3%( 1/44 ),respectively;and both patients with PHC had transfusion history.A partial nucleotide sequence(115~467nt)of the 5′ untranslated region(UTR) of HGV RNA positive products was cloned and sequenced by ABI Prism 377 DNA automatic sequencer.The homology of nucleotide sequence of these three HGV strains was 97.3% ~98.8%.Compared with GBVC(U36380),HGV(U44402)and genotype Ⅲ of HGV(D90601),its homology was 88.3%~89.4%,88.4%~89.4% and 95.2%~96.3%,respectively.Conclusion:There exist HGV infection in PHC and paid blood donor;suggesting PHC patients were probably infected HGV from transfusion,there may be no relationship between the pathogenesis of PHC and HGV.The three isolated HGV strains belong to the same genotype (HGV genotype Ⅲ).
