摘要
探究了喷他脒的合成工艺路线,以对羟基苯腈和1,5-二溴戊烷为起始原料合成了1,5-二-(4-氰基苯氧基)戊烷(Ⅰ);Ⅰ与干燥的氯化氢气体加成制得1,5-二-[4-(甲亚胺酸甲酯)苯氧基]戊烷盐酸盐(Ⅱ);Ⅱ的甲醇溶液中通入氨气,氨解合成了喷他脒(Ⅲ),总收率42.1%,其结构经1HNMR表征。
The pentamidine synthesis process route was explored,Pentamidine(Ⅲ) in total yield of 42.1% was synthesized in methanol ammonia by ammonolysis of dimethyl 4,4'-(pentane-1,5-diylbis(oxy))dibenzimidate dihydrochloride which was prepared using dry hydrogen chloride gas addition of 1,5-bis(4-cyanophenoxy) pentane produced by the reaction of p-hydroxybenzonitrile with 1,5-dibromopentane.The structure of Ⅲ was confirmed by1H NMR.
